Observations in rats and dogs by Wiebelhaus and colleagues (1) demonstrated that triamterene (SK&F No. 8542) (Figure 1) possessed antimineralo-corticoid activity. Furthermore, the site of such action appeared to be at the renal tubular level. Following 30-day toxicity studies by Saunders (2) in the same animal species at dose levels between five milligrams per kilogram of body weight and 40 milligrams per kilogram, which revealed no gross or microscopic changes in any organ system, it was decided to test the possible utility and toxicity of the agent in man. The following data are the results of these investigations.
