Adrenocorticotropic activity is not demonstrable in normal human plasma by current methods, but maximal stimulation of pituitary ACTH release (adrenalectomy, Addison's disease) results in measurable levels. When similar assay methods are applied to the plasma of patients with Cushing's disease (bilateral adrenal cortical hyperplasia), they fail to demonstrate adrenocorticotropic activity until complete adrenalectomy is performed. In order to implicate an adrenocorticotropin in the pathogenesis of this disorder, demonstration of increased activity in the plasma of the untreated patient is essential.
