It is well-recognized that the human adrenal gland synthesizes and secretes into the adrenal vein free androgens and sulfate derivatives. Recently the biosynthetic pathways of these androgens were established. "Normal," adenomatous, hyperplastic, and carcinomatous adrenal tissues removed at operation were incubated in buffer solution (1) alone, (2) with adrenocorticotrophic hormone (ACTH), (3) with tracers alone of H3-17α-rhydroxy-pregnenolone and C14-17α-hydroxyprogesterone, and (4) in equimolar amounts of (3). The data indicate that predominant biosynthetic Pathway I is 17α-hydroxypregnenolone → dehydroepiandrosterone → androstenedione. Although the Pathway II 17α-hydroxypregnenolone → 17α-hydroxyprogesterone → androstenedione was demonstrated, it is of lesser importance than Pathway I. It
