The intracellular site and mode of action of hypothalamic releasing factors for anterior pituitary hormones are unknown. Labeled (125I)-rabbit-antihuman growth hormone (HGH) was incubated with bovine anterior pituitary supernatant and shown on sucrose density gradient analysis to have bound to a specific polyribosomal (polysomal) peak. The bound radioactivity was liberated into the ribosomal monomer peak by pancreatic ribonuclease that also abolished the polysomes. Treatment with ovine hypothalamic extract containing growth-hormone releasing factor (SRF) resulted in a shift of radioactivity into a different (soluble) portion of the gradient. The latter treatment left the polysomes intact, indicating minimal contamination with pancreatic ribonuclease.
