The fact that penicillin resistance of staphylococci is mediated through the production of the enzyme penicillinase rather than by the resistance of a vital enzyme system has led to many attempts to inhibit penicillinase so as to allow penicillin to destroy the still susceptible staphylococci. The antipenicillinase sera of Housewright and Henry failed in this regard because intracellular penicillinase apparently was not reached and destroyed.
Total synthesis of penicillin by Sheehan and Henery-Logan suggested the possibility that substances could be synthesized that would have the antibacterial activity of penicillin but that would be resistant to the action of penicillinase.
