2,3-Diphosphoglycerate (DPG) is known to influence the oxygen equilibrium of hemoglobin and has been thought to participate in the regulation of glycolysis and nucleotide catabolism by inhibiting certain red cell enzymes. To examine further these properties of DPG in the intact red cell, fresh cells (hematocrit, 10%) were incubated with inosine, pyruvate, and phosphate (IPP), 20 mM each, for 16 hr at 37 C so as to increase their content of DPG.
