The amino acid composition of the isolated, normal canine glomerular basement membrane resembled that of tendon collagen but differed in being lower in glycine, proline, and hydroxyproline but higher in hydroxylysine. The hydroxylysine: hydroxyproline ratio in basement membrane was 0.42 compared with 0.07 in collagen. Carbohydrate makes up approximately 10% of basement membrane by weight, while collagen contains 0.6% carbohydrate. Basement membrane was solubilized by reduction of disulfide bonds in 8 M urea followed by alkylation.
