A method has been developed for studying lymphocyte function in various disease states, including uremia, neoplasia, macroglobinemia, autoimmune diseases, hyperglobulinemic conditions, transplantation, and others. The technique reproducibly measures incorporation of radioactive amino acid into trichloroacetic acid (TCA) precipitable counts by human peripheral lymphocytes in vitro. The technique in outline is as follows. Erythrocytes are sedimented from heparinized blood with gelatin, neutrophils adsorbed onto nylon, and lymphocytes washed and counted. Lymphocytes are cultured overnight in duplicate at 37 C in a carbon dioxide incubator in leucine-free media to which 14C leucine is added. No serum is required. After lysing the erythrocytes,
