To the editor: We have recently documented the occurrence of diffuse interstitial pulmonary fibrosis in a man after successful treatment of his Pneumocystis carinii pneumonia (1). Previous rat and human studies have also suggested that interstitial fibrosis may be a sequel of pentamidine isethionate-treated P. carinii pneumonia (2, 3). Is the fibrosis following pentamidine therapy drug related?
We have studied the effect of pentamidine on the rat lung in the absence of infectious disease in an attempt to answer this question. Fifty male, Sprague-Dawley, pathogen-free rats were divided into two groups, 10 controls and 40 test animals. Thirty-five rats were
